Severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2) is an RNA virus that causes coronavirus infection (COVID‐19). COVID‐19 is a highly contagious disease transmitted through respiratory droplets, saliva and other contact routes. Within 10 months of its outbreak, SARS‐CoV‐2 has infected more than 23 million people around the world. Evidence suggests that older adults are the most vulnerable to infection and have an increased risk of mortality. Reduced immunity and underlying medical conditions make them risk‐prone and vulnerable to critical care. Older adults affected with the SARS‐CoV‐2 virus present with distinct clinical manifestations necessitating specific treatment needs and management protocols. While it is crucial to prevent the spread of novel coronavirus (2019‐nCoV), the role of oral healthcare workers in addressing the specific needs of ageing adult patients by adopting specific guidelines and appropriate infection control protocols is timely. This paper aims to develop specific guidelines and protocols for the dental management of geriatric patients during the COVID‐19 pandemic.  相似文献

Dental tissue-derived stem cells (DSCs) provide an easy, accessible, relatively noninvasive promising source of adult stem cells (ASCs), which brought encouraging prospective for their clinical applications. DSCs provide a perfect opportunity to apply for a patient's own ASC, which poses a low risk of immune rejection. However, problems associated with the long-term culture of stem cells, including loss of proliferation and differentiation capacities, senescence, genetic instability, and the possibility of microbial contamination, make cell banking necessary. With the rapid development of advanced cryopreservation technology, various international DSC banks have been established for both research and clinical applications around the world. However, few studies have been published that provide step-by-step guidance on DSCs isolation and banking methods. The purpose of this review is to present protocols and technical details for all steps of cryopreserved DSCs, from donor selection, isolation, cryopreservation, to characterization and quality control. Here, the emphasis is on presenting practical principles in accordance with the available valid guidelines.  相似文献

The Antigen I/II (AgI/II) family of proteins are cell wall anchored adhesins expressed on the surface of oral streptococci. The AgI/II proteins interact with molecules on other bacteria, on the surface of host cells, and with salivary proteins. Streptococcus gordonii is a commensal bacterium, and one of the primary colonizers that initiate the formation of the oral biofilm. S. gordonii expresses two AgI/II proteins, SspA and SspB that are closely related. One of the domains of SspB, called the variable (V‐) domain, is significantly different from corresponding domains in SspA and all other AgI/II proteins. As a first step to elucidate the differences among these proteins, we have determined the crystal structure of the V‐domain from S. gordonii SspB at 2.3 Å resolution. The domain comprises a β‐supersandwich with a putative binding cleft stabilized by a metal ion. The overall structure of the SspB V‐domain is similar to the previously reported V‐domain of the Streptococcus mutans protein SpaP, despite their low sequence similarity. In spite of the conserved architecture of the binding cleft, the cavity is significantly smaller in SspB, which may provide clues about the difference in ligand specificity. We also verified that the metal in the binding cleft is a calcium ion, in concurrence with previous biological data. It was previously suggested that AgI/II V‐domains are carbohydrate binding. However, we tested that hypothesis by screening the SspB V‐domain for binding to over 400 glycoconjucates and found that the domain does not interact with any of the carbohydrates.  相似文献

The use of tooth cementum annulations for age determination has been deemed promising, exhibiting high correlations with chronological age. Despite its apparent potential, to date, the tooth cementum annulations method has been used rarely for estimating ages in archaeological populations. Here we examine the readability of cementum annulations and the consistency of age estimates using a sample of 116 adults from the Iron Gates Gorge Mesolithic/Neolithic series. Our examination of the method pointed to several sources of error that call into question the use of this method for estimating the chronological ages of archaeologically derived dental samples. The poor performance of the method in our analysis might be explained by taphonomic influences, including the effect of chemical and biological agents on dental microstructures. It is well known that interactions between epithelial components and mesenchymal components are essential for tooth development. Therefore, it has been postulated that both types of stem cells might be involved in the regeneration of dental hard tissues. Recently, mesenchymal dental pulp stem cells that have odontogenic potential were identified from human dental pulp. However, the existence of epithelial cells has never been reported in human dental pulp. In the present study, we isolated and characterized epithelial cell-like cells from human deciduous dental pulp. They had characteristic epithelial morphology and expressed epithelial markers. Moreover, they expressed epithelial stem cell-related genes such as ABCG2, Bmi-1, ΔNp63, and p75. Taken together, our findings suggest that epithelial stem cell-like cells might exist in human deciduous dental pulp and might play a role as an epithelial component for the repair or regeneration of teeth.  相似文献

Chemically cleaned and critical-point dried cells of a clonal culture were examined with scanning electron microscopy. Cells form filaments by valve-to-valve connections maintained by organic material which adheres to the central area of the valve face. Bending of filaments is probably restricted to some extent by the articulation of overlapping spatulate marginal spines with an adjacent underlapping set of much shorter spines (ridges), and with the mantle edge itself. Cell division results in three possible spine patterns for each cell: a set of overlapping and a set of underlapping spines; no overlapping sets of spines (two underlapping); or two sets of overlapping spines (no underlapping). Each filament inherits cells with spine set patterns in the ratio of 2 (with 1 set overlapping): 1 (with no sets overlapping): 1 (with 2 sets overlapping). Valvocopulae are shaped similarly to pleurae except that the partes exteriores of the valvocopulae are wider. The pars interior of both is delimited by an advalvar row of pores continuous around the cell apex. The pars exterior also has a row of pores, but it is median in the valvocopula and first pleura and does not continue around the cell apex. The valvocopulae always underlap the mantle and the pleurae always underlap their preceding band. The ends of both appeared attached, but may become free in acid-cleaned preparations. Bands alternate with each other so that the ends of the valvocopula attach to the first continuous apical portion of the first pleura; the ends of the first pleura attach in that same fashion to the second pleura but at the opposite apex; and all subsequent pleurae alternate in the same fashion with up to at least 13 pleurae/epicingulum. The continuous apical portion of each band is elevated so that a functional (but not structural) ligula is formed, with the continuous apical portion of alternate bands becoming adjacent and underlapping each other only in this region. The valvocopulae in a single cell, or of adjacent cells, may have their continuous apical ends on the same or on opposite apices. It is recommended that N. confervacea var. peregrina (W. Sm.) Grun. be merged with the nominate variety.  相似文献

New tools are available for teasing out aspects of life‐history variation among extinct species. Here we summarize research on the life histories of the extinct lemurs of Madagascar. There is a wide range of variation in dental developmental timing among these species, from among the most accelerated (Palaeopropithecus) to among the most prolonged (Hadropithecus) within the Order Primates. Rather than reflecting variation in body size, this diversity appears to relate to niche characteristics and encephalization.  相似文献

We report on the biometrics of cranial and dental features in 42 Macaca sylvanus specimens from various museums across Europe. Age classes were determined following dental criteria. Standard biometric landmarks were used to take 17 measurements on the cranium and five on the mandible. The permanent cheek dentition (except the whole C1/C1–P3/P3 complex) was also recorded with four measurements for each molar tooth (three for the P4/P4). Skull measurements show strong sexual dimorphism for characters tied to the mandibular and muzzle/facial portions. The unusual molar dimorphism of M. sylvanus is an unexpected result, because the Cercopithecoid molar pattern is often considered as generally conservative and not very dimorphic. A significant difference between M. sylvanus and other macaques also regards the absolute narrowing of the molar distal loph. This difference may result from a retained plesiomorphic condition, perhaps correlated with the less specialized and tougher diet maintained by M. sylvanus. A multivariate comparison for cranial (both sexual and ontogenetic) difference in M. sylvanus suggests that the full maturation of the male skull shape is delayed longer than what would be expected according to the most restrictive dental criteria adopted here for subadult/adult discrimination. The clustering results, indeed, indicate that significant phenetic differences persist between most of the male crania which are far advanced in their latest dental steps and those of the “fully developed adults”. Outcomes of final growth such as those observed in M. sylvanus suggest that polymorphic maturational patterns can mislead the assessment for a true adult skull shape.  相似文献

Periodontitis is a bacterially induced chronic inflammatory disease. Dental follicle progenitor cells (DFPCs) have been proposed as biological graft for periodontal regenerative therapies. The potential impact of bacterial toxins on DFPCs properties is still poorly understood. The aim of this study was to investigate whether DFPCs are able to sense and respond to lipopolysaccharide (LPS) from Porphyromonas gingivalis, a major periopathogenic bacterium. Specifically, we hypothesized that LPS could influence the migratory capacity and IL‐6 secretion of DFPCs. DFPCs properties were compared to bone marrow mesenchymal stem cells (BMSCs), a well‐studied class of adult stem cells. The analysis by flow cytometry indicated that DFPCs, similar to BMSCs, expressed low levels of both toll‐like receptor (TLR) 2 and 4. The TLR4 mRNA expression was down‐regulated in response to LPS in both cell populations, while on protein level TLR4 was significantly up‐regulated on BMSCs. The TLR2 expression was not influenced by the LPS treatment in both DFPCs and BMSCs. The migratory efficacy of LPS‐treated DFPCs was evaluated by in vitro scratch wound assays and found to be significantly increased. Furthermore, we assayed the secretion of interleukin‐6 (IL‐6), a potent stimulator of cell migration. Interestingly, the levels of IL‐6 secretion of DFPCs and BMSCs remained unchanged after the LPS treatment. Taken together, these results suggest that DFPCs are able to sense and respond to P. gingivalis LPS. Our study provides new insights into understanding the physiological role of dental‐derived progenitor cells in sites of periodontal infection.  相似文献
